Should the chain tightness be measured while the bike is on a stand or on the ground?

Whether it's on the stand or on the ground, it doesn't matter in this case, as in EITHER case the suspension should be unloaded.
If your unitrack/swingarm is worn out to the point that putting the bike on a stand gives you another couple inches of movement as opposed to it being on the sidestand, you have far more serious problems than chain tension.
This is not a distinction without a difference.

The manual says (9-4) 'Support the mototcycle on its side stand.'
Unless you have something wrong setup-wise with your free sag, unless you have something wrong with your suspension, when the bike is on the side stand, the suspension will 99% of the time be completely stretched out..unloaded..topped out.
...which is no different than it would be on a stand.
If your idea of lowering the bike means having a 3" free sag as a result of your shock spring adjustment...I can't help 'ya.
